Methylation analysis of 5S rRNA genes by the methylation-sensitive HpaII (H) restriction enzyme and its methylation-insensitive MspI (M) isoschizomere. The left two panels show Southern blot hybridisation with the 5S rDNA probe; the right panel (EtBr) shows ethidium bromide-stained DNA fragments before the hybridisation. The probe hybridised with a ~200 bp MspI fragment corresponding to a monomer. Hybridisation signals with the HpaII fragments were of high molecular weight. Samples “f” and “b” were fin and blood isolates, respectively. The samples from fin tissue were slightly degraded which probably accounted for smear signals
