Establishment of a mouse model with persistent HBV infection and liver fibrosis. The mice were injected with AAV-HBV vector (2 × 1011 viral genome equivalents (vg)) via the tail vein. The control mice were injected with the same volume of PBS. Then, serum was collected at 1, 3 and 6-months post injection. The parameters associated with persistent infection and liver fibrosis were analyzed. a–d The expression levels of α-SMA, TGF-β, Collagen I and Collagen III were assessed using quantitative RT-PCR in the liver tissue of HBV (−) and HBV (+) mice. e–g The concentration of Hydroxyproline, TGF-β and TIMP1 in the serum was assessed by ELISA. h The liver tissues were collected at 1, 3 and 6-montha post injection. Masson’s stain and Sirus red staining were performed to assess collagen deposition. Significantly increased fibrosis was found in the HBV (+) mouse. Representative images of Sirius red (upper panels) and Masson’s staining (lower panels) of liver sections at the indicated time points are shown. Statistical analyses were performed using a one-way analysis of variance. The data represent the mean ± SD (n = 4). Compared to HBV (−) mouse:* P < 0.05, ** P < 0.01,*** P < 0.001
