Approximately equal amounts of WT BCG Danish and selected knockouts were mixed and grown in media, infected into PBMC derived bovine macrophages, and inoculated into cattle lymph nodes. Inoculants and recovered BCG were enumerated on selective media. Cattle lymph nodes were resected after 3 weeks emulsified and BCG enumerated on selective media. Significance of the in vivo survival between of the three knockouts and the WT and control were all p < 0.001 using Tukey’s test. Error bars represent standard errors
